    • This project will allow the identification of drug-like compounds able to inhibit TMBIM4, which could constitute potential drugs and mechanistic tools for cell biology studies. The project will also enable the identification and validation of
      additional ER or GA Ca2+ channels that affect glioma progression that will be further explored as therapeutic targets. This exploratory project will pave the way for subsequent lines of research in pharmacology and drug delivery systems against glioma invasion.

      • The Blue4Skin project intends to use sardine co-products, from the company “Conservas Pinhais & Cia” in order to be used as raw materials in innovative health products for the skin, with added value. The optimization of extraction, fractionation, isolation and identification of marine natural products and the corresponding screening study of various bioactivities related to skin health is important to enable the use of these residues and co-products and ensure the safety, quality, and acceptability of the product. It is also intended to contribute to the mitigation and adaptation to climate change through the reduction of waste from these industries. Thus, the exploitation of marine biomass and recovery of sea co-products seems to be a promising alternative to sustainable uses of marine resources with greater economic benefits, in line with the concepts of circular economy and blue bioeconomy.

        • The project explores existing multi-omics datasets associated with powerful bioinformatic tools to identify novel molecular targets associated with α- to β-cell-like transdifferentiation. In combination with multigenomic data from small molecule polyphenol metabolites (SMPM) studies, the project aims to facilitate the advancement of these small molecules into lead candidates for nutraceutical and pharmaceutical applications in diabetes management.

        • Non-communicable diseases (NCDs) are among the most pressing global public health challenges. Cardiovascular diseases (CVD) are the leading cause of death worldwide, while metabolic disorders such as obesity and type 2 diabetes mellitus (T2DM) significantly reduce quality-adjusted life years (QUALYs). Adopting healthy lifestyles—particularly regarding nutrition and physical activity - is critical for disease prevention and health promotion across the lifespan. However, the real-world impact of such interventions in at-risk populations remains underexplored. This pilot project aims to evaluate the effects of simple lifestyle changes - focusing on diet quality and physical activity - on cardiovascular and metabolic risk markers in sedentary adults.
          The study will recruit 60 omnivorous men and women over 40 years old, presenting with overweight or obesity and a sedentary lifestyle. Participants will undergo comprehensive baseline assessments, including body composition via dual-energy X-ray absorptiometry (DXA), anthropometric measurements, dietary intake using the Diet Quality Index-International (DQI-I), and physical activity levels via the International Physical Activity Questionnaire (IPAQ). Fasting capillary blood samples will be used to assess key metabolic biomarkers. Hemodynamic and vascular health will be evaluated non-invasively through ankle-brachial index (ABI) calculations and assessments of lower limb perfusion asymmetries. Risk stratification will include the SCORE2® algorithm to estimate 10-year cardiovascular risk and the FINDRISC tool for T2DM risk. Participants will be grouped accordingly at baseline.
          The intervention will involve small dietary modifications aligned with Portuguese national guidelines, alongside a minimally supervised home-based physical activity program designed by our team, following the FITT (Frequency, Intensity, Time, and Type) principle. The program is intended for daily completion at each participant's convenience. Data will be collected at baseline and at 1, 3, and 6 months. The primary goal is to improve metabolic and cardiovascular profiles, ultimately reducing long-term CVD and T2DM risk. This project will provide evidence that accessible, consistent lifestyle changes can meaningfully improve health and well-being.

            • This project explores how plants from Portuguese-speaking regions could provide new treatments for wound healing and breast cancer. By investigating Lysil oxidase enzymes and their role in these conditions, the project aims to find natural compounds that can modulate LOX activity. These compounds will be tested on breast cancer cells and skin cells to understand their effects. Additionally, the project will use advanced techniques to enhance the effectiveness and safety of these natural compounds. This collaborative effort brings together experts from various fields to train young researchers and establish international partnerships for future progress.

                • Multidrug resistance (MDR) is characterized by complex resistance mechanisms that protect cells from compounds with different chemical groups and modes of action. It is prevalent in all forms of cancer, one of the greatest health challenges present in modern society, as well as in drug-resistant bacteria and fungi.In this project, we intend to target the primary mediators of MDR in cancer (ABC transporter protein family) through a novel therapeutic approach and mechanism, recently discovered in the PI group.

                  Within the ABC family, there are three main transporters responsible for MDR in cancer: P-gp/ABCB1, MRP1/ABCC1 and BCRP/ABCG2. However, multidrug-resistant bacteria
                  (MDRB) is becoming also a serious problem. In 2019, antimicrobial resistance killed more people than HIV or malaria and antimicrobial resistance is predicted to cause 10 million deaths by 2050 (cancer: 8.2 M). Therefore a completely new paradigm to tackle MDR mediated by ABC transporters is needed and is proposed herein.

                  Despite the large evolution in knowledge involving MDR related efflux, no compound passed clinical trials into general use. Therefore, the search for efficacious and safe P-gp inhibitors is urgently needed. However, signs of real clinical progress in using ABC transporters as pharmacological targets were already reached:

                  In 2012, ivacaftor (VX-770) was approved for therapy in cystic fibrosis patients with the G551D mutation on ABCC7;
                  more recently, in 2021, encequidar (HM30181A) was discovered as the first-in-class intestine specific P-glycoprotein Inhibitor.
                  The team as been studying ABC transporters with great success to understand the detailsregarding the efflux mechanism and to understand how and where allocrites and inhibitors interact with the transporters, and the details about allocrite recognition and signal propagation leading to efflux. Moreover, Dr. Daniel dos Santos group recently identified and characterized two allosteric drug binding sites (aDBSs) by means of in silico fragment- based molecular dynamics and tested a small library of thioxanthone and flavanone derivatives. Several compounds were experimentally assayed and identified to be able to decrease the verapamil-stimulated ATPase activity.

                  The ABC-MDR-REVERSAL project proposes to rationally discover and design novel ABC modulators acting through this novel allosteric mechanism.

                  • Rare dermatological conditions like Xeroderma pigmentosum (XP) are autosomal recessive syndromes resulting from specific congenital defects in the nucleotide excision repair pathway. There is no known cure for these disorders, which are life-threatening and cause a significant impact in the life of these patients. Despite presenting variable symptoms, common dermatological manifestations can be identified, such as heightened sun sensitivity, freckling in sun-exposed areas of the skin and dryness.

                    Presently, the management of these clinical conditions is contingent upon their severity, with the primary focus on early diagnosis, lifelong protection from UV light, and symptom-based supportive treatments. In accordance with current medical guidelines, patients are advised to apply sunscreens with high SPF daily to minimize UV exposure.

                    In the case of XP, a pronounced increase in the incidence of skin cancer has been reported and the patients undergo frequent surgical resection of skin lesions. The current challenges of the management of these skin conditions are mainly related to safety concerns regarding daily exposure to titanium dioxide nanoparticles used as photoprotective agents in sunscreens, as well as regarding the topical application of anticancer drugs.

                    The NANO4RARESKIN project aims to upgrade the management of rare skin conditions related to DNA repair disorders by designing daily maintenance, multifunctional and nanotechnology-based formulations with photoprotective, antioxidant, and skin barrier recovery properties.

                    The strategy for innovation will be based on the optimization of lipid-based nanosystems containing ceramides and phyto-antioxidants and further combined with titanium dioxide nanoparticles using a Quality-by-Design approach. These nanosystems will be characterized and incorporated in semisolid formulations, finally proceeding to stability, safety, and efficacy assessments.

                    Upon completion of the NANO4RARESKIN project, it is expected that at least one nanotechnology-based formulation will be poised for knowledge transfer and have potential to make a significant contribution towards a safer and cost-effective daily management of rare skin conditions.

                    The accomplishment of the ambitious objectives of NANO4RARESKIN will be ensured by the know-how on lipid-based nanosystems for skin drug delivery and by the strategic collaboration with a multidisciplinary team of CBIOS researchers. This befitting group will also be dedicated to dissemination and communication activities, so that the major breakthroughs of the project become available for the scientific community, industry, patients’ associations, and general society.

                    • The general objective of the REDALIM-MIC Network is to constitute an interdisciplinary network of mobility and experimentation that, through its knowledge and the contribution and commitment of the participating companies, allows the identification of compounds with antimicrobial activity present in food plant species of regional interest, which makes it possible formulate functional foods that contribute to preventing and combating severe infectious processes, both acute and chronic. At the same time, this proposal aims to highlight the role that food and healthy eating play, not only from a nutritional point of view but also as a source of bioactive compounds that play an important role in terms of prevention and/or treatment of certain pathologies. The preventive role of functional foods will be highlighted by strengthening the immune system, producing defenses against infectious agents,

                      The proposal is based on a regional collaborative research project with the participation of companies and cooperatives in the region. It is relevant to highlight that we intend to work with foods of regional interest, that is, foods that grow or are cultivated in the Ibero-American region, which are marketed in our territory, giving added value to our internal production.
