Multidrug resistance (MDR) is characterized by complex resistance mechanisms that protect cells from compounds with different chemical groups and modes of action. It is prevalent in all forms of cancer, one of the greatest health challenges present in modern society, as well as in drug-resistant bacteria and fungi.In this project, we intend to target the primary mediators of MDR in cancer (ABC transporter protein family) through a novel therapeutic approach and mechanism, recently discovered in the PI group.
Within the ABC family, there are three main transporters responsible for MDR in cancer: P-gp/ABCB1, MRP1/ABCC1 and BCRP/ABCG2. However, multidrug-resistant bacteria (MDRB) is becoming also a serious problem. In 2019, antimicrobial resistance killed more people than HIV or malaria and antimicrobial resistance is predicted to cause 10 million deaths by 2050 (cancer: 8.2 M). Therefore a completely new paradigm to tackle MDR mediated by ABC transporters is needed and is proposed herein.
Despite the large evolution in knowledge involving MDR related efflux, no compound passed clinical trials into general use. Therefore, the search for efficacious and safe P-gp inhibitors is urgently needed. However, signs of real clinical progress in using ABC transporters as pharmacological targets were already reached:
In 2012, ivacaftor (VX-770) was approved for therapy in cystic fibrosis patients with the G551D mutation on ABCC7; more recently, in 2021, encequidar (HM30181A) was discovered as the first-in-class intestine specific P-glycoprotein Inhibitor.
The team as been studying ABC transporters with great success to understand the detailsregarding the efflux mechanism and to understand how and where allocrites and inhibitors interact with the transporters, and the details about allocrite recognition and signal propagation leading to efflux. Moreover, Dr. Daniel dos Santos group recently identified and characterized two allosteric drug binding sites (aDBSs) by means of in silico fragment- based molecular dynamics and tested a small library of thioxanthone and flavanone derivatives. Several compounds were experimentally assayed and identified to be able to decrease the verapamil-stimulated ATPase activity.
The ABC-MDR-REVERSAL project proposes to rationally discover and design novel ABC modulators acting through this novel allosteric mechanism.

Nanosolutions to upgrade the daily management of rare skin conditions

Rare dermatological conditions like Xeroderma pigmentosum (XP) are autosomal recessive syndromes resulting from specific congenital defects in the nucleotide excision repair pathway. There is no known cure for these disorders, which are life-threatening and cause a significant impact in the life of these patients. Despite presenting variable symptoms, common skin manifestations can be identified, such as heightened sun sensitivity, freckling in sun-exposed areas of the skin and dryness.
The management of these clinical conditions is contingent upon their severity, with the primary focus on early diagnosis, lifelong protection from UV light, and symptom-based supportive treatments. In accordance with current medical guidelines, patients are advised to apply sunscreens with high SPF daily to minimize UV exposure.
In the case of XP, a pronounced increase in the incidence of skin cancer has been reported and the patients undergo frequent surgical resection of skin lesions. The current chalenges of the management of these skin conditions are mainly related to safety concerns regarding daily exposure to titanium dioxide nanoparticles used as photoprotective agents in sunscreens, as wel as regarding the topical application of anticancer drugs.
The NANO4RARESKIN project aims to upgrade the management of rare skin conditions related to DNA repair disorders by designing daily maintenance, multifunctional and nanotechnology-based formulations with photoprotective, antioxidant, and skin barrier recovery properties.
The strategy for innovation wi l be based on the optimization of lipid-based nanosystems containing ceramides and phyto-antioxidants and further combined with titanium dioxide nanoparticles using a Quality-by-Design approach. These nanosystems wi l be characterized and incorporated in semisolid formulations, finaly proceeding to stability, safety, and efficacy assessments.
Upon completion of the NANO4RARESKIN project, it is expected that at least one nanotechnology-based formulation wi l be poised for knowledge transfer and contribute towards a safer and cost-effective daily management of XP. The accomplishment of the NANO4RARESKIN objectives wi l be ensured by the know-how on lipid-based nanosystems for skin drug delivery, supported by a multidisciplinary team of CBIOS researchers. This befitting group wi l also be dedicated to dissemination/communication activities, so that the major project breakthroughs may have societal impact.

Portugal's seascapes and immersive media: representations of Nazaré from early 20th century stereoscopic photographs to early 21st century 360º videos
Portugal’s Seascapes & Immersive Media: Nazaré Immersive is a transdisciplinary exploratory project supported by ULHT-CICANT that aims to develop and test immersive experiences of stretches of the Portuguese coast, based in this specific case on the collection of stereoscopic photographs by Álvaro Laborinho dated from the beginning of the 20th century and belonging to the collection of Dr. Joaquim Manso Museum in Nazaré.
From the re-mediation of Álvaro Laborinho's stereoscopic photographs - in the sense of Bolton & Grusin (1998) - we propose the production of new analogue and digital contents, as well as the respective supports and exhibition materials, namely:
a 360º virtual reality (VR) video an interactive application for the 360º video; a website to support the project; a dome for shared VR experiences; several exhibition materials: printed panoramic screens, exhibition supports, printed stereoscopic cards for stereoscopic viewers.

NewsArcade@Classroom emerges as a natural extension of the ongoing CREA project, "NewsArcade-Seriously Play the News!" (101060250), which introduced an Authoring Tool designed to gamify news stories and bolster young adults’ media literacy skills, fostering a deeper engagement with traditional media. Building upon this foundation, NewsArcade@Classroom endeavors to elevate the existing NewsArcade format to serve the specific needs of media and information literacy education. Leveraging the capabilities of the existing tool, the project seeks to create new educational formats that will be used by educators with students for media and information literacy education purposes.
NewsArcade@Classroom thus endeavors to engage students aged 12-25 in critical thinking, digital literacy, and media and information analysis by offering interactive and immersive learning experiences, as well as cultivating communities of news-literate youth across diverse European contexts. Our vision is to cultivate a generation of digitally literate and media-savvy individuals who are equipped to navigate and contribute meaningfully to the media landscape, thus protecting democratic values. We envision a future where playful media literacy education is integral to the curriculum, empowering students to engage thoughtfully with media content and participate actively in civic discourse.
In addition, the project aims to support and empower educators in delivering effective media literacy instruction. By providing educators with innovative tools and resources for gamified news storytelling through NewsArcade, the project seeks to enhance their capacity to engage and inspire students in media and information literacy. Finally, through a cross-border collaborative effort of partners and their networks, the project wishes to share knowledge and exchange best practices between different regions and stakeholders of Europe, contributing to a more cohesive and interconnected media literacy landscape.
No Barriers aims to research and facilitate the inclusion of individuals with intellectual disabilities in multi-sectoral employment, focusing on the development and implementation of social inclusion policies. Recognizing that disability contributes to social exclusion, the project seeks to address barriers that prevent intellectually disabled individuals from engaging in social relations, cultural and social activities, education, accessing basic services, and participating in the economic field.

While most research on digital cultures tends to focus on online and connected practices, a fast-expanding field of scholarship explores how people disconnect from one or more forms of digital media (Lomborg & Ytre-Arne, 2021). This exploratory study seeks to advance digital media scholarship, and to expand the scholarship on the nascent field of digital disconnection to the still underexplored context of Portugal, as most existing research in this area is predicated in Northern European countries. It proposes to do so by looking at disconnection as “embodied and affectively experienced” (Coleman & Paasonen, 2020, p. 1) across different aspects of everyday life or social situations. Our study aims to illuminate the complexities of people’s engagement with and disengagement from digital media, by focusing on micro-environments of everyday situations characterized by their affective intensity, vulnerability, and liminality. Particularly, the exploratory study posits the question: how are online and offline articulated in situations such as pilgrimage, mourning, everyday parenting, or activism with digital media? We approach the everyday encounters with digital media as atmospheres (Sumartojo & Pink, 2018), ephemeral elements of our everyday experiences and environments, that encompass the sensorial as well as affective modes of engagement. Our methodological approach is ethnography, fit to grasp the affective and sensorial complexities of situations such as pilgrimage, mourning, everyday parenting, and activisms.
The reason for the existence of art is supra race, language, religion and nationality. Its service to humanity is the symbol and legacy of multilateralism and cultural diversity. Art, while increasing our awareness of human values, points to what we need most in this pandemic period. If there is one thing that can save humanity, it is universal unity. The reason for the existence of art does not depend solely on aesthetic values. On the one hand, it is an ointment to the deepest wounds of our souls, on the other hand, it shakes our judgments and dogmas. Artistic practices that bring people together, relax, inspire and hope during the epidemic process, which includes concerns and uncertainties caused by Covid-19, will form the basis of our project activities.
PandemiArt intends to deal with traditional art practices and applications of digital art from an inclusive perspective. The team aims to create artistic products with digital content by taking art from an innovative and contemporary perspective as a means of free expression of the individual. The idea of art and art education will play an active role in creating visual awareness, positively drawing attention to the effects and protective measures of Covid-19 under social responsibility
Considering the social responsibility duty of educational institutions, the idea of creating a transnational awareness against the pandemic has reinforced the partnership that is in the heart of the PandemiArt project. Project partners accommodate different types of institutions which will ensure that education is handled in a wide-ranging manner.

PORTSIGN (Portuguese Sign Language Dataset and Recognition Project) aims to develop an open, multimodal dataset for Portuguese Sign Language (LGP) alongside an ethical and inclusive artificial intelligence model capable of accurately recognising and interpreting sign language gestures. Combining computer vision, deep learning and sign language linguistics, the project seeks to enhance communication accessibility and promote the digital inclusion of the Deaf community. Through a participatory approach involving Deaf educators, native signers and interpreters, PORTSIGN will collect, annotate and validate high-quality gesture data, develop hybrid AI recognition models, and demonstrate their application in educational and communication settings. The resulting open-access dataset will serve as a benchmark resource for research in sign language technologies, artificial intelligence and inclusive digital solutions, while strengthening collaboration between the scientific and Deaf communities and fostering more accessible human–machine interaction.
